霍亂毒素

由霍亂弧菌產生的霍亂毒素（CT）是一種多聚體腸毒素，可將ADP-核糖轉移至G蛋白，將腺苷酸環(huán)化酶鎖定在“開啟”位置。

霍亂毒素如何引起腹瀉？通過與腸細胞膜結合，霍亂毒素刺激細胞腺苷酸環(huán)化酶系統(tǒng)，引起氯離子和碳酸氫根離子的過度分泌，這導致增加的液體分泌和霍亂疾病的嚴重腹瀉特征。CT的五聚體B亞基與GM1單唾液酸神經(jīng)節(jié)苷脂細胞膜受體結合，存在于許多細胞類型中，允許其在細胞培養(yǎng)中實驗性使用。結合后，毒素被內吞并逆行進入內質網(wǎng)，其中酶活性A亞基（CTA1）易位至胞質溶膠。在胞質溶膠中，CTA1催化Gs蛋白的ADP-核糖基化，其激活腺苷酸環(huán)化酶，并因此增加cAMP的細胞內濃度。
霍亂毒素如何用于疫苗佐劑開發(fā)？ 與疫苗佐劑的開發(fā)相關，CT有效地調節(jié)哺乳動物免疫系統(tǒng)并充當共同施用的抗原的佐劑。
